We have completed a screening of over 20 single, dual, and quad expression systems in two cell lines. We have identified several potential candidates for further study.
Work is currently being written up for publication.
Not at this time. After publication, this project will be closed and we will pursue further funding from federal and private grantmaking organizations.

Stress tolerance experiments with the transgenes have revealed protective effects in one cell type and we are proceeding to test additional cell types. Further, we have developed and successfully engineered cells with the multicistronic expression vectors, resulting in expression of pairs of the genes in a single human cell.
Continue collecting stress tolerance data, including with the multicistronic expression vectors.

We have validated expression and localization of the engineered transgenes in human cells, the designed proteins localize to the targeted organelles correctly. We have performed preliminary stress tolerance experiments in cancer cells. We are currently expanding the stress tolerance results and analyzing the data.
Based on our data analysis we will select lead combinations for multicistronic expression.

Several things have moved forward. Work completed prior to the award has been published and is available online at: https://www.nature.com/articles/s41598-024-62693-w/ We have designed localization sequences for our original four targets (nucleus, endoplasmic reticulum/sarcoplasmic reticulum, plasma membrane) and additionally designed a “humanized” mitochondrial variant, replacing the tardigrade mitochondrial targeting sequence with a human version. The genes have been synthesized, cloned into appropriate vectors, and transformed into bacterial stocks. The genes have been transposed into human cancer cells.
We are currently working towards validating expression and localization. Following that, we will proceed with stress tolerance testing of the human cells.
